Apoptotic Effects of an Anti-carcinogenic Kalopanaxsaponin A on the Growth of Trichomonas vaginalis

Abstract
Kalopanax pictus Nakai (Araliaceae) is one of the herbs used in oriental medicine to treat neuralgia, rheumatic arthritis, lumbago, furuncle, carbuncle, wound, diarrhea and scabies. Kalopanaxsaponin A, purified from cortex of Kalopanax pictus is known as an anti-carcinogenic agent is also used for the treatment of various types of vaginitis. We performed to examine the effects of kalopanaxsaponin A on the inhibition of growth and fine-structure of Trichomonas vaginalis (T. vaginalis). Kalopanaxsaponin A dose-dependently reduced the viability of T. vaginalis. In the electron microscopy study, we also observed the ultramicroscopical changes of T. vaginalis following the treatment with kalopanaxsaponin A.
Anti-protozoan effect was great when treated with 16¥ìg/ml kalopanaxsaponin A after 1 hour treatment.
There was no statistically significant difference in production of nitrite comparing with control group. We evaluated the anti-protozoan effect of kalopanaxsaponin A against T. vaginalis by electron microscopy. SEM shows that in T. vaginalis cultivated with TYM medium treated with 4¥ìg/ml for 1hour, at first, motor organ, especially axon was shrinked and flagella began to disappear, and induced a destruction of cell membrane in T. vaginalis. TEM shows that the vacuoles in cytoplasm were larger, hydrogenosomes were smaller, and the number of polyribosome was decreased in T. vaginalis treated with 4¥ìg/ml for 2 hours. By staining of DAPI, the degenerative change was evaluated a fragmentation of nucleus and budding appearance of nucleus, a typical pattern of apoptosis in some T. vaginalis. Therefore, our results
show that the kalopanaxsaponin A induced an anti-protozoan activity through apoptosis in T. vaginalis. (Cancer Prev Res 12, 210-219, 2007)
